Culture of a Guatemalan strain of Polyporus umbellatus Fr. in solid media and production of sclerotia In vitro

The myeelia characteristics of a native strain of Polyporus umbellaius were described in different culture media and temperatures. The strain (P. umbellaius 25.2000) was reactivated in PDA agar and incubated at 26 0 C for 22 days. The radial grow th rate was determined in culture media SAB, PDA and PDA-IM EMA al temperatures of 18 and 26°C. The most vigorous and rapid growth was observed in the PDA medium-lM at 24 C at 10 days of incubation with an average diameter of 37.70 mm and a radial growth rate of 3.77 mm / day. For the inoculums production it were used grain sorghum, wheat, barley. which were incubated at 18 and 26 ° C. Growth was evaluated for 50 days, a period in which no mycelial grow th was observed in any of the evaluated substrates at both temperatures. Sclerotia production was evaluated during four months, incubating (lie substrate (oak sawdust) at 18 ° C. After this period there was no formation of sclerotia despite of the complete colonization of the substrate. Therefore, it was concluded (hat the conditions evaluated in this study were not appropriate for the production of sclerotia.
