Abstract

The work consisted of analyzing, using the Random Amplification of Polymorphic DNA (RAPDs) technique, the kinship relationships in a population of Triatoma dimidiata (Reduvidae, Triatominae) collected from a single human dwelling over the course of a year. The selected dwelling was located in the village of El Cuje, Municipality of Pueblo Nuevo Viñas, Department of Santa Rosa. This area is considered a highly endemic area for Chagas disease in Guatemala. 130 T dimidiata individuals were collected, from which leg DNA was extracted and amplified using short primers or oligonucleotides L1, L4, and H3 from the Operan company. The amplification results allowed the analysis of 14 dominant loci with allele frequencies between 0.1 and 0.6. The number of "full-sib" families (siblings from the same male and female parent) was estimated at 17. The number of individuals in each family and the number of families reflected moderate inbreeding and demonstrated intra-household gene flow. Six families were found to be made up of three or more full siblings, and 11 families with two or fewer siblings from the same father and mother. These 11 families may indicate external gene flow and/or polyandry. These results are extremely useful for control purposes, since a population with high inbreeding will be easily controlled by traditional methods. This is not the case for a population showing high diversification. In the case of T. dimidiata from Pueblo Nuevo Viñas, inbreeding is moderate, indicating a species in the process of diversification. These results reveal that control strategies for T. dimidiata must take into account the population movements of the species and the degree of inbreeding in each population.
