Abstract

The document summarizes the tests carried out for the standardization of a serological method (Enzyme-linked immunosorbent assay, ELISA) for the diagnosis of amoebiasis, using antigen obtained from axenic culture of Entamoeba histolytica. The study was carried out in two phases, the first consisted of obtaining antigen; for this, the amoebas were harvested in culture flasks in the logarithmic phase of growth, thus obtaining a solution containing 5 ug/ul of amoebic antigen. In the second phase, the sensitivity and specificity of the antigen obtained was tested by confronting it with monoclonal and polyclonal antibodies by means of the ELISA technique. It was found that under these conditions the antigen was capable of detecting antibody titers up to 1:6400, using commercial conjugates at double the recommended dilution. The antigen was also tested against patient sera and commercial controls, and was simultaneously compared to a commercial ELISA method, where the same results were obtained by both methods. It is concluded that the antigen prepared in the laboratory gives good results. It is recommended that more tests be carried out in order to standardize the method for diagnostic purposes and seroepidemiological studies that provide a real framework for the prevalence of amoebiasis in Guatemala.
