Comparative study between two methods to quantify guaifenesin in simple expectorant syrup

In the present investigation, the method of quantification of Guaifenesin in simple syrup by Thin Layer Chromatography and UV Spectrophotometry was developed and validated, in order to have an alternative analysis technique that would be useful in laboratories that did not have a Liquid Chromatograph. For this purpose, eight guaifenesin standard solutions were made in a concentration range of 40 to 58 ug/mL to check Beer's law. For this, Linear Regression was applied and a 95% confidence interval was used. Likewise, the TLC/UV Spectrophotometry technique was compared with the official USP 2-3 analysis technique (HPLC), by means of the quantification of the active ingredient in a pilot batch of known concentration, performing 20 determinations per technique. For statistical analysis, paired t* and descriptive statistics were applied. In the results of the quantification of the pilot batch of guaifenesin syrup, by the HPLC method, a mean of 102.51 mg/5 mL was found, with two standard deviations of 4.38 and a coefficient of variation of 2.14%. In the TLC/UV Spectrometry method, a mean of 100.27 mg/t mL was obtained, two standard deviations of 6.94 and a coefficient of variation of 3.46%. A mean of the differences of 2.44 and a standard deviation of 3.07, a paired tM of 3.55 with a theoretical t” of 2.09 were also obtained. The results indicate that the TLC/UV Spectrometry technique allows the identification and quantification of guaifenesin in syrup with acceptable precision (a CV less than 100%). e 5%). However, this technique does not replace the HPLC method in terms of precision or accuracy. It is recommended to apply the TLC UV spectrum technique to other pharmaceutical forms of guaifenesin, as well as to other active ingredients.Downloads
